【中文摘要】 从桃红平菇单核菌丝分离出原生质体铺于含0.6mol/l蔗糖的CM表面,经紫外线处理一定时间后,培养7—10天后长出的菌落被转于MM之上,在其上生长十分缓慢或停止的菌落通过各种营养添加物测其营养需求,最后得到一个需要补充丙氨酸和天冬氨酸才能正常生长的双缺突变体。试验表明,该突变体能很好地分离出原生质体,且在含0.6mol/l蔗糖的MM上不能再生,因而具有较好的应用前景。
【英文摘要】 Protoplasts isolated from monokaryotic mycelia of Pleurotus salmonea-straminens were spread on CM containing 0.6 mol/l sucrose and irradiated by U. V. After 7—10 days, the regenerated colonies were transplanted on MM and those showing little growth were detected further with various kind of nutritional requirement., One auxotrophic mutant, double deficient in Ala and Asp, was obtained at last; from which a good protoplast yield could be obtained. It will be useful in protoplast fusion study because its protoplasts can not regenerate on MM containing 0.6 mol/l sucrose.
