【中文摘要】 探讨用超声提取法提取桦褐孔菌中桦褐孔菌多糖的工艺条件及桦褐孔菌多糖对肝癌SMMC7721细胞株的抗增殖作用。设计正交实验,对桦褐孔菌的超声提取工艺进行研究,并用MTT比色法研究桦褐孔菌多糖对肝癌SMMC7721细胞株的抗增殖作用。桦褐孔菌多糖超声提取的最佳工艺条件为95℃、15倍水、20kHz、超声60min,桦褐孔菌多糖在1.0 ̄16.0μg/mL范围内对肝癌SMMC7721细胞株的增殖均有抑制作用,且表现出浓度相关性。
【英文摘要】 OBJECTIVE To investigate the optimal purified protocol of extracting Inonotus obliquus polysaccharide using ultrasonication and the effect of the extractions from Inonotus obliquus polysaccharide on anti-proliferation for SMMC7721 cells of liver cancer.METHORD The method of orthogonal analysis was used to extract Inonotus obliquus polysaccharid with ultrasonication,The methods of MTT was used to study the anti-proliferative effect of the polysaccharide on the hepatocarcinoma SMMC7721cells.RESULT The best condition used in the course of extraction of Inonotus obliquus polysaccharide was: the temperature was 95℃,the volume of water was 15 times,the frequency of ultrasonication on the extraction was 20kHz,the optimal duration of extraction was 60min.The Inonotus obliquus polysaccharide had the inhibiting action on SMMC7721 cells of liver cancer line in the concentration of 1.016.0ug/mL by means of MTT assay,and exhibited a strong dependence on concentration.
