【中文摘要】 以苯酚硫酸法作为灵芝多糖的测定方法,研究了从固体发酵灵芝菌丝体中提取水溶性多糖的工艺,对浸提过程中的温度、时间、固液比及提取次数4个因素进行了单因素试验和正交试验,并利用SephadexG-100凝胶色谱法和薄层层析法对主要组分GLPⅠ进行了分子量测定和单糖分析。结果表明,灵芝多糖的最佳提取条件为浸提温度95℃,浸提时间40min,固液比1∶20和提取次数为2次;GLPⅠ分子量约为179900,初步判断为甘露聚糖。
【英文摘要】 Using phenol-sulphuric acid method to detect the contents of polysaccharides, the extraction of the water-soluble polysaccharides from Ganoderma lucidum mycelia was studied. The optimum extraction conditions were determined by single-factor test and orthogonal test. And the molecular weight and monosaccharide composition of GLPⅠ were determined by Sephadex G-100 gel filtration chromatography and thin-layer chromatography. The results showed that the optimum extracting temperature, time, ratio of mycelia to water ( w∶v ) and extracting times were 95℃, 40 min, 1∶20 and 2 times. The molecular weight of GLPⅠ was approximately 179 900. GLP I was probably composed of D-mannose.
