【中文摘要】 本文比较了双核菌丝制备的香菇原生质体在不同培养基上的再生率,观察了原生质体再生过程.以存活率37%的剂量进行紫外线诱变处理后,选出102份再生克隆,从中选出在CM培养基和木屑培养基上菌丝长速较快的材料23份,进行栽培试验,其中10份产量提高,出菇较早.在随后的二次或三次组织分离后代的栽培试验中,大部分材料保持了增产、早熟的性状,经显著性测验.其中2份材料的产量显著提高.本研究初步建立了用原生质体诱变技术选育香菇菌株的试验程序.
【英文摘要】 The regeneration efficiency of Lentinula edodes protoplast prepared from dikaryon mycelium was compared on different media and the regeneration procedure was also observed under inverted microscope. 23 clones with more rapid mycelium growth rate on CM medium and sawdust medium were selected from 102 regenerated clones after being treated by UV radiation at D37 dosage. The cultivation experiment showed that 10 of the 23 regenerated clones expressed the characters of yield increment and early fruitbody formation, and that most of the progeny obtained through 2 or 3 times of tissure separation kept the characters of yield increment and early maturity and the yield of 2 regenerated clones increased significantly according to the significance test. In addition, the experiment procedure for selecting L, edodes strains with protoplast technique was also established in this study.
