【中文摘要】 利用有机溶剂萃取法提取树舌灵芝MP-01发酵液中灵芝酸,并用分光光度法测定提取液中灵芝酸的含量,确定了分光光度法测定的最佳比色条件为:比色波长为520 nm,5%香草醛-冰醋酸和高氯酸用量分别为0.2 mL和1.0 mL(在反应系统中浓度分别为1.0%和83%),显色温度70℃,显色时间为25 min,在此条件下测得醇溶物和醇沉物提取液中灵芝酸含量分别为0.022 mg/g和0.049 mg/g。
【英文摘要】 Ganoderic acid produced during the growth of Ganoderma MP-01 in submerged culture was extracted using ethanol and chloroform,and its concentration was determined spectrophotometrically at 520 nm using oleonolic acid as the standard.Optimal assay conditions were: vanillin concentration 1%,perchloric acid concentration 83%,reaction temperature 70 ℃;reaction time 25 min.Ganoderic acid concentrations in SIE(substance insoluble in ethanol) and SSE(substance soluble in ethanol) fractions were 0.049 mg/g and 0.022 mg/g,respectively.
