【中文摘要】 应用RAPD技术对13个蜜环菌(Ar millaria mellea)菌株进行了遗传多样性分析。结果表明,在供试的60条RAPD随机引物中,有16条可扩增出清晰、稳定的DNA条带;聚类分析表明,以黄绿蜜环菌(Ar millaria luteo-virens)为外类群,在相似系数0.760水平时,可将供试的13个菌株分成四大类:第一大类为A.m0010、洋县M-8、蜜0903、A.m0005、宁强A9-1、蜜环菌A9和A.m0006,第二大类为A.m0007、A.m0004和A.m0008;第三大类为蜜金乡和蜜三明;第四大类为A.m0001。研究结果表明RAPD技术可以有效区分蜜环菌菌株并分析其遗传多样性。
【英文摘要】 Genetic diversity among thirteen Armillaria mellea strains was analyzed by RAPD using sixteen primers selected from 60 tested primers that displayed specific and stable amplification patterns.A dendrogram constructed using the amplification data and Armillaria luteo-virens as the out-group separated the test strains into four groups at a coefficient of 0.760.Group I consisted of strains A.m0010,Yangxian M-8,Mi 0903,A.m0005,Ningqiang A9-1,Mihuanjun A9 and A.m 0006,Group II of strains A.m0007,A.m0004 and A.m0008,Group III of strains Mi Jinxiang and Mi Sanming,and Group IV of strain A.m0001.Our data showed that RAPD was a suitable method for differentiating the test strains and was effective in reflecting genetic diversity.
