【中文摘要】 对不同形态猪苓(Polyporus umbellata)菌核(猪屎苓、铁蛋猪屎苓和鸡屎苓)的rDNA(18S r DNA,ITS1-5.8S rDNA-ITS2)和β-tub1进行扩增、测序。序列分析表明:所得rDNA和β-tub1序列完全相同。以Neighbor-joining(N-J)方法构建ITS(ITS1-5.8S rDNA-ITS2)序列系统发育树,供试菌株同GenBank中收录猪苓(P.umbellata)类聚于同一分枝且序列完全相同,表明不同形态菌核的猪苓之间亲缘关系较近。
【英文摘要】 Genetic relationships between three Polyporus umbellata strains,designated Zhushiling,Tiedanzhushiling and Jishiling on the basis of differences in sclerotium morphology,were analysed using rDNA(18S rDNA,ITS1-5.8S rDNA-ITS2) and β-tub1 fragment sequences.Although the test strains exhibited dissimilar sclerotial features,no differences were detected in their rDNA and β-tub1 sequences.Using the neighbor-joining method,a phylogenetic tree was constructed on the basis of the ITS data and other ITS sequences deposited in GenBank.This revealed a relatively high genetic similarity between the test strains,another P.umbellata strain and several Grifola umbellata strains.
