【摘要】 液体培养蛹虫草(Cordyceps militaris)鲁山株C0511,以菌丝体产量为指标,对培养时间、接种量、温度和培养基起始pH进行单因素试验,结果表明,最佳培养时间为96h,接种量为10%,适宜温度为22~26℃,pH为5.8。正交试验结果表明,影响蛹虫草菌丝液体培养的主次因素依次为培养温度、培养时间、接种量和pH。鲁山菌株C0511液体菌种按10%的接种量接入起始pH为7.0的液体培养基中,25℃培养96h菌丝产量为30.13g/L。
【Abstract】 Monofactorial testing revealed that mycelial yields of Cordyceps militaris (strain Lu Shan C0511) grown in submerged culture were maximal under the following conditions:culture temperature 2226 ℃,incubation time 96 h,inoculum size 10% (v/v),medium pH 5.8. Application of orthogonal testing revealed that mycelial yields were influenced predominantly by culture temperature,followed by incubation time,inoculum size,and the initial pH of the medium. When 10% (v/v) liquid spawn of strain Lu Shan C0511 was inoculated into medium adjusted to pH 7.0 and incubated at 25 ℃ for 96 h,the mycelium yield was 30.13 g/L.
