【摘要】 通过IgE诱导大鼠被动皮肤过敏反应(PCA)实验、细胞毒性试验、IgE诱导肥大细胞组胺释放、钙摄入量实验、Western blot试验,来研究桦褐孔菌(Inonotus oblipuus)乙醇提取物EEIO的抗过敏作用。试验发现,EEIO明显抑制了大鼠皮肤伊文思蓝渗出,对IgE诱导肥大细胞的细胞内钙摄入、组胺以及肿瘤坏死因子-α、白细胞介素6的释放有剂量依赖抑制作用。表明,桦褐孔菌乙醇提取物具有一定的抗过敏作用。
【Abstract】 We have studied the anti-allergic effects of an orally-administered ethanolic extract of Inonotus obliquus(EEIO) by evaluating anti-DNP IgE-mediated passive cutaneous anaphylaxis(PCA) in rats,EEIO cytotoxicity,histamine release,calcium uptake,and TNF-α and IL-6 secretion by IgE-induced rat peritoneal mast cells(RPMCs) incubated with EEIO.EEIO(1 100 μg/mL) had no adverse effect on RPMC viability under the test conditions,and significantly inhibited Evans blue leakage in antigen-challenged rats sensitized by anti-DNP IgE.EEIO also suppressed antigen-stimulated calcium uptake and histamine release in RPMCs,and Western blot analysis revealed that EEIO inhibited the antigen-stimulated expression of TNF-α and IL-6 in RPMCs in a dose-dependent manner.Our data indicate that EEIO exhibits anti-allergic effects.
