【摘要】 采用组织分离法从市售牛肝菌子实体分离得到两株性状稳定的分离株Lq4-2和Be2425,利用ITS序列分析并结合分离子实体的形态特征对两分离株进行鉴定,并对其基础生物学特性进行了研究。分离株Lq4-2鉴定为牛肝菌科(Boletaceae)成员,分离株Be2425鉴定为广义美味牛肝菌(Boletus eduliss.l.);两分离株能在PDA培养基和MRD合成培养基上良好生长;菌丝生长的适宜温度为2528℃,适宜pH为56,最佳氮源为酒石酸铵;Lq4-2的最佳碳源为葡萄糖,而Be2425的最佳碳源为果糖和葡萄糖。
【Abstract】 Two bolete strains,Lq4-2 and Be2425,were isolated using standard tissue culture methodology from fruit bodies purchased at two local markets,and assigned to the Boletaceae and Boletus edulis sensu lato,respectively based on internal transcribed spacer(ITS1-5.8S-ITS2) sequence analysis.Both strains exhibited highest mycelial growth rates and growth vigor on potato dextrose agar,although several other media also supported fungal growth.Both strains grew optimally between 2528 ℃,between pH 56,and with ammonium tartrate as the N source.Among several carbon sources tested,glucose supported the highest Lq4-2 growth rates,and either fructose or glucose in the case of Be2425.
