【摘要】 构建了香菇135菌株(Lentinula edodes)菌丝生物量与其DNA含量的标准曲线。通过一系列的筛选工作,确定了标准菌丝的培养条件及DNA提取系统,得到了三个不同培养批次香菇135菌丝体构建的高重现性的关联标准曲线,它们均具有较高的相关系数(>0.99),统计构建标准曲线的所有90个数据,测定的单点误差范围为0.05%13.74%(<14%)。所得数据表明在操作范围内,菌丝生物量与DNA含量之间存在很好的线性关系,每mg香菇135标准培养菌丝(干重)对应的DNA含量为6.1μg。
【Abstract】 Standard curves relating DNA content and mycelial biomass were constructed for Lentinula edodes,strain 135.Adopting a set of standardized mycelium growth conditions and DNA extraction procedures,standard curves relating DNA concentration with mycelial biomass for three separate batches of L.edodes,strain 135,exibited a correlation coefficient >0.99.The error range for a total of 90 determinations(18 samples,five replicates of each sample)was between 0.05%-13.74%.Our data indicated that,under ...
