【摘要】 以蝙蝠蛾拟青霉(Paecilomyces hepiali)菌丝体为材料,以腺苷和D-甘露醇为评价指标,在单一酶提取实验基础上,采用正交实验设计对复合酶法提取工艺进行优化。优化的复合酶法提取工艺:浸提pH 5.5、浸提温度40℃、酶组合为纤维素酶0.1%+蜗牛酶0.1%,在此工艺条件下,提取物中甘露醇、腺苷含量分别为12.56%和2 065μg/g。
【Abstract】 Factors affecting the enzymatic extraction of D-mannitol and adenosine from Paecilomyces hepiali mycelium were optimized using orthogonal tests designed on the basis of single factor experiments.The data revealed that the optimal extraction pH,temperature and enzyme combination values were pH 5.5,40 ℃,and 0.1% cellulase and 0.1% snailase,respectively.Maximal levels of mannitol and adenosine obtained under optimal extraction conditions were 12.56% and 2065 μg/g,respectively.
