【中文摘要】 香菇(Lentinusedodes)菌株的DNA片段通过克隆转移到质粒载体BluescriptM13—(pBS)以获得部分基因文库。文库中三个随机克隆和一个密环菌中rDNA克隆被用作限制性片段长度多态性探针。其中三个探针可检测到多态性,揭示了32个野生和栽培香菇菌林的遗传分离。菌林间的相似性用平均链锁聚类方式(UPGMA)聚类,相似性超过60%的分为一组。23个菌株被分成4组。9个菌林与其它菌株相似性均低于60%而独立存在。从我们的结果看出,香菇的遗传分离普遍存在于菌株间而与地理位置没有相关性。RFLP具有研究香菇种内遗传分离的潜力,并可用作遗传分析的遗传标记,因此能够为育种工作和遗传分析提供科学依据。
【英文摘要】 EcoR Ⅰ restriction fragment of genemic DNA from the cultivated mushroom,Lentinus edodes,was cloned into the plasmid vector Bluescript MI 3 (pBS). Three randomclones from the genemic library and one rDNA clone of Armillaria melles were used to probefor restriction fragment length polymorphisms(RFLP).Three of the four probes detectedpolymorphisms and were used to demonstrate genetic divergence and detected polymorphismsin 32 wild and cultivated strains of Lentinus mushroom,The similarities between strains werecalculated using average linkage clustering (UPGMA),Based on 60 percent of similarity,23 strains were separated into four groups and anotlier 9 strains were not clustered intoany group due to their less similarity,From our resultS,the genetic divergence occurs amoungstrains but do not have a connection with geographic position,These results suggest thatRFLP have potentiaI to study interspecific genetic divergence in L.edodes and that RFLPcould be served as genetic markers for formal...更多 genetic analysis and thereby provide thefundation for breeding programmes. 还原
