【中文摘要】 研究了不同酶种类、酶浓度、酶解温度和时间、渗透压稳定剂、pH值及菌龄等因素对云芝原生质体制备以及不同再生培养基对云芝原生质体再生的影响。结果表明,菌龄3d的云芝菌丝体用3%溶壁酶,以pH5.5,0.4mol/L的KCl为稳定剂,30℃酶解5h获得的原生质体产量最高;用RM1再生培养基培养,原生质体再生率最高。
【英文摘要】 We have examined the effects of selected parameters(nature and concentration of lytic enzyme,exposure time,temperature,nature,concentration and pH of osmotic stabilizer,mycelial age) on Coriolus versicolor protoplast production.Highest protoplast yields were obtained from 3-day-old mycelium after treatment with 3%(w/v) lywallzyme for 5 h at 30 ℃ using 0.4 mol/L KCl(pH 5.5) as osmotic stabilizer.Of six media tested,highest mycelium regeneration rates were obtained on RM1 medium consisting of 20%(w/v) potato,1%(w/v) yeast,0.3%(w/v) KH2PO4,0.15%(w/v) MgSO4,0.003%(w/v) Vitamin B1,0.4 mol/L KCl and 3%(w/v) agar.
