【摘要】 采用小鼠S180移植瘤实验研究了巴西菇子实体中麦角甾醇的体内抗肿瘤活性,并通过巨噬细胞吞噬功能测定(中性红法)、脾淋巴细胞增殖功能检测(MTT法)、体外抑杀肿瘤细胞(MTT法)以及鸡胚绒毛尿囊膜(CAM)等试验初步探讨了麦角甾醇抗肿瘤活性的作用机理。结果表明,巴西菇子实体中提取的麦角甾醇在剂量10 mg.kg-1.d-1、50mg.kg-1.d-1和100 mg.kg-1.d-1时的瘤重抑制率分别为62.75%、76.61%和79.18%,说明麦角甾醇有较强的抑瘤活性。在巨噬细胞吞噬试验和脾淋巴细胞增殖试验中,麦角甾醇各剂量组与阴性对照组相比无统计学差异(p>0.05),没有剂量依赖趋势,说明其对巨噬细胞吞噬功能和脾淋巴细胞增殖均无明显促进作用,可能不是通过增强免疫来发挥抑瘤活性的;麦角甾醇体外对人胃癌细胞SGC-7901和肝癌细胞Bel-7402的抑制试验中,与对照组相比均无统计学差异(p>0.05),说明其抑瘤活性也不是通过细胞毒作用来发挥的;在CAM实验中,麦角甾醇与相同剂量(5μg.mL-1)的阳性对照地塞米松相比,表现出了更强的抑制血管生长的作用(抑制率63.46...
