【摘要】 利用DEAE-Sepharose Fast Flow离子柱层析和Sephacryl凝胶柱层析对灰树花(Grifola frondosa)子实体水提醇沉粗多糖进行分离纯化,对分离纯化到的一种多糖GFP75-2-2B进行了分析。紫外光谱鉴定结果表明,GFP75-2-2B不含蛋白质、核酸和酚类成分;离子色谱分析表明其单糖组成为岩藻糖、鼠李糖、半乳糖、葡萄糖和甘露糖,摩尔比为1.0∶1.3∶3.6∶4.0∶4.5。GFP75-2-2B对巨噬细胞株RAW264.7进行体外免疫活性的研究结果表明,GFP75-2-2B有明显刺激巨噬细胞分泌NO的活性。
【Abstract】 A water-soluble,crude polysaccharide preparation was obtained from Grifola frondosa fruit bodies by hot water(100 ℃) extraction followed by ethanol(75%) precipitation.A purified polysaccharide,GFP75-2-2B,was obtained from the crude material by successive fractionation using DEAE-Sepharose Fast Flow ion exchange chromatography and Sephacryl S gel filtration chromatography.UV spectroscopy detected no protein or nucleic acid in GFP75-2-2B,which consisted of fucose,rhamnose,galactose,glucose and man...
